不同种质怀地黄生育期根际土壤挥发性有机物变化分析

摘要:
【目的】分析不同种质怀地黄生育期根际土壤挥发性有机物(VOCs)的变化特征,为探讨地黄连作障碍及其与根际土壤中化学成分变化的关系提供理论依据。【方法】以金九、北京1号、白选3个种质地黄为研究对象,于07-28(地上部位快速生长期)、08-24(地下块根膨大初期)和10-18(成熟期)采集根际土壤样品,分别编号为J-7、J-8、J-10,B-7、B-8、B-10和X-7、X-8、X-10,利用气相-质谱联用(GC-MS)技术检测其乙酸乙酯和二氯甲烷提取物的VOCs,通过面积归一化法对各成分的相对含量进行计算,并应用统计学方法对不同种质地黄各生育期根际土壤VOCs进行主成分分析(PCA)及聚类分析(HCA),利用正交偏最小二乘法判别分析(OPLS-DA)结合t检验筛选各生育期差异的VOCs。【结果】从3个种质地黄不同生育期根际土壤乙酸乙酯提取物中共得到56个共有色谱峰,从二氯甲烷提取物中共得到50个共有峰。不同时期地黄根际土壤VOCs差异较为明显,其中8月根际土壤VOCs与7月、10月差异更为明显,3个生育期地黄根际土壤乙酸乙酯提取物与二氯甲烷提取物共有差异VOCs分属于酮类、醇类、酚类。【结论】不同种质地黄同一时期根际土壤VOCs表现基本一致,但同一种质地黄根际土壤VOCs在不同生育期存在差异。

Variation of volatile organic compounds in rhizosphere soil during growth period of Rehmannia glutinosa from different germplasms

Abstract:
【Objective】This study analyzed the variation characteristics of volatile organic compounds (VOCs) in rhizosphere soil during the growth period of Rehmannia glutinosa from different germplasms to provide basis for exploring the relationship between continuous cropping obstacles of R. glutinosa and chemical composition changes in rhizosphere soil.【Method】Three germplasms of R. glutinosa including Jinjiu,Beijing No.1,and Baixuan were selected and rhizosphere soil samples were collected on July 28 (the rapid growth period),August 24 (the early stage of underground tuber enlargement) and October 18 (the maturity period).Then,VOCs in ethyl acetate extract and dichloromethane extract were detected by GC-MS and the normalization method was used to calculate relative percentage content of each component.Finally,principal component analysis (PCA) and cluster analysis (HCA) were performed on VOCs,and orthogonal partial least squares method (OPLS-DA) combined with t-test was used to screen different components.【Result】A total of 56 common chromatographic peaks were detected in ethyl acetate extract of the R. glutinosa rhizosphere soil during the growth period,and 50 common peaks were detected in dichloromethane extract.VOCs in rhizosphere soil of R. glutinosa at different periods had obvious differences.Among them,VOCs in rhizosphere soil of R. glutinosa in August had larger differences than those in July and October.The shared differential VOCs in ethyl acetate extracts and dichloromethane extracts of R. glutinosa rhizosphere soil at the three growth periods belonged to ketones,alcohols,and phenols.【Conclusion】The VOCs in R. glutinosa rhizosphere soils of different varieties were basically the same in same periods,but VOCs in same varieties were different in different growth periods.
